Most likely to be left after some updates or temp files after downloads. See if it's associated with some program , if not - you can delete it.
I agree with BisVal.
It may have been used as a temporary cache while an update or download was installed. Try opening the folder and if there's nothing in it just delete it if it's bothering you.
The Windows Malicious Software Removal tool sometimes leaves these scattered in the root of your drives. As Seavixen said delete it if it is empty.
Thanks to you all. I have been curious about those for a long time. I have been hesistant to delete files ever since the early 1990's when I deleted my DOS folder to free up some space. I didn't know a PC could come to a halt quite so quick.

I started with Windows 3.1 and never did live down the fact I deleted all the program icons from the desktop because it looked untidy!
My boss didn't speak to me for a week. Happy days.

These files in the root of the system drive are normally temporary files relating to updates to windows defender and the like. they should be deleted as part of the update process.
It should be OK to delete them but to be safe I would move them somewhere, (in documents?), and run for a day or so to make sure that nothing breaks
