Try right clicking on it and selecting Run as Administrator, see if that helps.
Otherwise, fill out your System Specs in User CP and provide a link to where you downloaded the BIOS update from. | My System Specs | | System Manufacturer/Model Number Dell Precision 370 OS Windows 7 Pro X64 CPU Intel Pentium 4 Dual LP 3.4Ghz Memory 4GB DDR PC2-5200 ECC Graphics Card NVIDIA Quadro FX 3400/4400 Sound Card SoundMAX Integrated Digital Audio Monitor(s) Displays HP 22" w2207 LCD Screen Resolution 1680 x 1050 Hard Drives 300GB Maxtor 6L300RD PATA

Some motherboards now lock the bios so it can not be flashed. To check this, go to the Motherboard web site, or reboot and open the bios menu and see if there is a setting to lock or unlock, or otherwise protect the bios.
Cheers!

Ok, what type of motherboard (or type of pc) is it?
And have you checked physical connections inside your pc? The optical drive connections could be loose.
If you feel uncomfortable with that, take it to a pc repair place. | My System Specs | | System Manufacturer/Model Number ATX Custom Build 2012 OS Windows 7 Enterprise x64 SP1 CPU Intel Core i3-2100 @ 3.10GHz Motherboard Intel DH67CL desktop ATX Memory 10 GB DDR3 Graphics Card Asus EAH5450 series (Radeon) Monitor(s) Displays Samsung SyncMaster 226BW, Samsung Syncmaster P2450H Screen Resolution 226BW: 1680 x 1050 & P2450H: 1920 x 1080 Keyboard HP Wireless Elite Keyboard Wireless Keyboard Mouse Microsoft Touch PSU Cooler Master Extreme Power Plus 500W Case Thermaltake V3 Black Edition Cooling stock cooling with added intake fan Hard Drives Western Digital 320 GB sata (boot), Samsung 640 GB sata, Seagate 2 TB sata (data) Internet Speed Comcast Cable business class <=18Mb {averages 12Mb} Other Info Optical drives: LG SuperMulti Blue, HP DVD 1260T

either, but I believe a loose connection is more probable.
But...given you inability to flash the bios, the mobo could be bad.
How old is it? Under a year, then perhaps under warranty | My System Specs | | System Manufacturer/Model Number ATX Custom Build 2012 OS Windows 7 Enterprise x64 SP1 CPU Intel Core i3-2100 @ 3.10GHz Motherboard Intel DH67CL desktop ATX Memory 10 GB DDR3 Graphics Card Asus EAH5450 series (Radeon) Monitor(s) Displays Samsung SyncMaster 226BW, Samsung Syncmaster P2450H Screen Resolution 226BW: 1680 x 1050 & P2450H: 1920 x 1080 Keyboard HP Wireless Elite Keyboard Wireless Keyboard Mouse Microsoft Touch PSU Cooler Master Extreme Power Plus 500W Case Thermaltake V3 Black Edition Cooling stock cooling with added intake fan Hard Drives Western Digital 320 GB sata (boot), Samsung 640 GB sata, Seagate 2 TB sata (data) Internet Speed Comcast Cable business class <=18Mb {averages 12Mb} Other Info Optical drives: LG SuperMulti Blue, HP DVD 1260T
