yes, but it will only work on the computer that you are setting this up on.

Go to Control Panel > Administrative Tools > Computer Management > Disk Management

Select the USB drive in the graphical layout. right click and go to Change Drive letter

Assign it a drive letter and click OK. Now when you plug it into the computer, it will show up with that drive letter. or it should....

This won't carry over to other computers.

Shortcut: type in "Disk Management" in the start menu search bar and select "Create and format hard disk partitions"

plug in USB drive--go to Computer Management>Disk Management--right click the disk - select 'Change drive letter and paths' -the drive should be highlighted--select 'change' - assign drive letter - click OK and OK....done!

If you want the USB stick to retain the drive letter on other PC's 1st of all select a seldom used drive letter...say from the middle of the alphabet down...
Then after assigning it in Disk Management also give the stik a Volume Label...it can be any name you want.
This will make it retain the drive letter on any PC...the only time it will change is if it's used on a PC that already uses that drive letter.
