We need to know what you are using.

    PS2 Mouse and Kayboard?
    USB Mouse and Keyboard?
    Wireless Mouse and Keyboard?

    Have you restarted the computer?

Try swapping ports. Typically the Mouse Port is green and the keyboard is pink or orange. Also the mouse port is typically the inboard port. with the computer up and running, plug in USB mouse and keyboard.

Try a system restore, some corruptiion may have ocurred.

If that does not work, go the borrowed keyboard and mouse route.

Similar trouble. New system running Windows 7 x64 on asus maximusIII Formula. Will randomly come out of sleep mode with inoperative keyboard. reinstalling keyboard drivers by unplugging (USB) and plugging back in results in keyboard working again. Power options set to: HD off after 20min,sleep after 30 min, hybrid sleep off, Hibernate never, wake timers enable, USB selective suspend enabled power button action shut down (Should this be different if I push power button to awaken?). Using an old PS/2 keyboard seems to avoid the problem.Have downloaded newest drivers for the keyboard from logitech.Suggestions? Thanks.
