What brand of mouse do you have?
Look in Control panel and click on mouse and check settings.

Are you sure you didn't first click the volume icon? Because the mouse wheel by default controls the volume slider when the slider is open.

Main - Windows 7 Pro SP1 64-Bit; 2nd - Windows Server 2008 R2

Check the other suggestions first, but there is a program named Volumouse - control the sound volume with a wheel mouse that will do that. Do you have it installed?
