I've googled the heck out of this and due to the common keywords of the search, you can imagine that my results were just a huge mix of unrelated items.
Can anyone advise if with having a fully up and running version of Win 7 x64 Home Premium I can use a full DVD of Win7 x64 Ultimate to upgrade? Or would I have to do a full wipe and start all over again?

Anytime upgrade only adds-on the Programs & Features from version to version.

You can use a full version key as well as upgrade version to Anytime Upgrade from HP to Ultimate. Just type Anytime into Start Search box to begin the process which takes about 20 minutes tops.
But keep in mind that Home Premium may have everything you need so using a full version (or even upgrade) Ultimate key might be a significant waste of money, when you already own HP on that machine (if OEM) for life and can use full Ultimate anywhere also for life.
I would save a Win7 backup image externally so you can fall back to HP at any time in the future if you decide you want to use Ultimate elsewhere. Then after Anytime save an Ultimate backup image so you dont have to do both again.
