If you are trying to Upgrade XP to Win7.
You must initiate the install as a "Upgrade Install' you will see in step #8:
8. Click on the Upgrade option. (See screenshot below)
NOTE: Choose Custom if you want to do a clean install and completely replace your current operating system.
This is where you choose 'Custom Install', from there it will format the partition and perform a Clean install.

If you are trying to do a Custom or Clean install from a Full Version and the DVD won't boot, and as mentioned your BIOS is set to Boot from DVD and the original copy won't Boot, Is this correct?
If yes, next thing to check is if your DVD drive is functioning correctly. Test it with another boot CD/DVD as suggested by sup3rsprt.

Has this DVD been used to install Win7 before?

Insert it while the OS is running and see if you can read anything from it.
