How to create partition in 2nd HDD?

Dear Sir,

I am using 160GB HDD with 4 partition which is connect on SATA 0 port. Now I want to connect one more 500GB HDD on SATA 1 port & want to make 2 partition under windows for 250GB of each. Kindly tell me is it right & also give me complete procedure because i am doing this job first time.
