How do I enable my front microphone and headphone jack with Windows 7?

The front panel should automatically be enabled, providing the cable for the front panel audio is connected to the motherboard, and you are using the correct sound driver. I assume you have Realtek as I used to have a P8Z77 LE Plus motherboard and it had Realtek on board audio.
On many PC cases there are two connections available for front panel audio, "HD Audio" and "AC 97". The HD Audio is preferred as it as some jack sensing functions that the older AC97 does not have.
I looked at the Realtek HD Audio Manager on my system and there is no option to enable or disable the front panel jacks. There is an option to mute or not mute the speakers when a headset is plugged in but that's all.
Even if you only have the generic, basic function, Windows installed High Definition Audio drivers installed instead of the correct hardware drivers you should still get front panel headphone operation.
Several possibilities for the front panel. (1) Cable not connected (2) defective cable (3) defective front panel assembly in case.
Your motherboard does have Realtek. You can download the latest HD Audio driver direct from Realtek. www.realtek.com
I'd go with number 3. I recently installed a 5.25 inch internal card reader front bay onto my custom built PC with an Intel D101GGC board using onboard Realtek ALC861 audio chip. the card reader bay drive also had additional USB 2.0 ports and HD audio & Microphone jacks along with the cables (including the 8-wire HD audio cable that connected perfectly onto the front panel HD audio connector on the Intel board). I removed the old AC97 audio cable and the old built in front panel module with old USB, mic & headphone jacks since I don't use them anymore. With the new card reader bay drive installed and the cables connected, the Realtek control panel app correctly detected the front panel ports and anytime I connect the headphones onto the new HD audio jack, the rear speakers are turned off and they turn back on when I unplug the headphones.
