VMs, Paragon Alignment Tool and Defrag

I recently installed the Paragon Alignment Tool, which supports virtual machines. I tested it in an XP VM and it completed the alignment without problems. I'm wondering if defragging the VM file from the Host OS will undue the alignment? And regarding defragging a VM, is it better to run it inside the VM first and the host OS second? TIA
