syngus21, I wanted to give you some feedback on setting my Nikon Coolscan 4000 up, followed your procedure and it worked like a charm, even the slide loader works and I'm plenty stoked. Much thanks!!

Sygnus21, Thank you very much for having done this Edit. I have your original file copied everywhere I can, but wanted a fresh one since my 4000ED showed an odd error message. I was just about to uninstall the whole driver and revised your original (saved) post ...until I happened to read this twice, and felt a click inside:
Simply, I Updated and Browsed again. Clicked Next, got an O.K. message. Turned On the Coolscan, and the error message had disappeared.
Still, I don't know why did it stop working, and what did the error message meant. Nor if this issue was previously treated in this Topic thread.
Now, ...now I will see if the Coolscan is in fact working as it did before ....

1- Yes, I have it working now and hope it will do for a very long time from now. Followed steps 7, 8, 9 and the error message didn't show up again.
The yellow triangle never appeared.
I had uninstalled VueScan last Wednesday, but this did not prevent the error message to show up any time I launched NikonScan. I have it working now, but didn't install VueScan again.
2- The question / issue is: what may have happened ? - it was working like a charm, I took my summer holidays and stopped scanning for four weeks, and all of a sudden I got an error message when launching NikonScan only (VueScan worked fine, but since I prefer NikonScan I uninstalled VueScan but, as said, this didn't heal the problem).
3- By chance, before reinstalling the whole Nikon driver, I experimented with steps 7, 8, 9 only, and this procedure proved enough to make the scanner to work as nice as before. But I still doubt about installing VueScan again, because I have not a clue on what was the cause that made Windows 7 to lose contact with some part of the Nikon driver.
Did I make myself clear enough? English is not my mother language, thus it's very probable that I posed my question / issue in a rather confusing way. Sorry about that, I always try to write the best I can.
Last, I want to thank you for having updated the link to download your file ..
I've no idea what happened. I have both Nikon and Vuscan installed and have never experienced that issue, nor seen it reported here. In fact I just now tried the scanner for the first time in about 8 months and the software and everything still works.
One thing to check is that you don't have a loose connection between the the scanner (USB/FireWire) and PC as that could cause a driver issue. Also check that the power cord isn't loose. Other than that I can't say.
Anyway you say it's working now so I'd go from there.

Mmmmm .... I missed to mention it, but I did so since it was part of your original post. I didn't notice it could be loose, but anyway I unplugged and made myself sure to plug it tight.
If a loose FW connection can cause odd driver issues (not a simple 'device not found'), you may have nailed on the answer, and at the same time, underlined an instruction line I had understated (it seemed such an obvious thing ... ). Thank you again. I will install VueScan with more confidence now.

A loose connection can very well cause a driver issue just a loose connection can cause a light to fail. In the case of the driver this can happen if it's rapidly being cycled off and on by a loose connection. This activity can cause the driver to corrupt itself and not function properly.
Sometimes you'll see this happen to the OS (Windows) and/or drivers when there is a rapid or sudden shutdown and startup of the system. It usually take more that one time for this to happen, though once is enough.
