You cast it at the animal/monster/creature, and try to kill then before the spell ends. If you do, you capture their souls. Make sure you have soul gems in your inventory. Also Some souls are too big for some souls gems, you are gonna need bigger soul gems.

Oh i see thats the way.I used to cast spell after creatures/animals died
