Currently, I have Windows 8 Dev Preview on my desktop, but if I were to put Windows 7 Home Premium back on it, would it be able to use all my RAM? Or would I need to get the professional edition? Today I upgraded to 12 GB of RAM.
You'll be fine. Home Premium 64-bit can address up to 16GB. I believe the Home Basic version is limited to 8GB...
win 7 premium x64 can handle up to 16gb of ram. Unless you need more then you need pro or ultimate

The usable memory may be less than the installed memory on Windows 7-based computers
I have somewhat of a similar question. My new build will have Win 7 Home Premium 64bit with 16gb of RAM. If I find I need more RAM, and I use the Anytime Upgrade to Pro, will I be able to take advantage of the extra RAM? Or will I have to purchase the full version?

The anytime upgrade downloads the files needed to upgrade your system from Home Premium to Professional, and on completion your system will be running with Windows 7 Pro, meaning you will be able to benefit from extra RAM should you decide to install it.
So, no, you won't have to purchase the full version.
However, as you're building a new computer, why don't you just buy Windows 7 Pro in the first place? It'll save you the bother of upgrading.
