Running two antimalware at the same time might degrade system performanceMalware antimalware 1.46,windows defender
Do you have a firewall?
The free windows firewall is a good choice
My personal preference is malwarebytes in lieu of your antimalware software, but to answer your question the answer is yes.
You do not want more than one malware product, however. Defender is anti malware.
You may want to look into the free WinPatrol. Scotty, the cute liitle dog, will bark and notify if something new is added to your system. A nice device to have. I know that our own Corrine, recommends the product, too.

You common sense is your best defense. You will get a lot of opinions but.... my 2 cents is, Use Microsoft Security Essentials and Malwarebytes. This will probably free up some of you resources and along with your common sense you should be in good shape.

It's not so much about installing an anti virus or whatever and just forgetting about it. You should check sure that it updates itself, but more importantly--
Make sure that your web browser and any plug-ins (EX: Adobe Flash / Reader, etc, etc) are always up to date, as well as installing all important Window's update when they are released. That will make the life of your anti virus much easier as it won't have to deal with 0-day flaws so often (They'd be patched.)
