MSE is good for a free AV, but even Microsoft does not recommend it over paid-for AV software. Sure it's better than AVG and McAfee, but its not in quite the same league as Bitdefender, Trendmicro, Norton and the like. Thats why we always recommend using it with MBAM or Hitman Pro, and having a decent firewall.
Running Windows 7 Ultimate x64 with UAC Maxed.

Malwarebytes, SuperAntiSpyware, SpyBot S&D.
Main Anti-Virus Protection - Kaspersky Pure or ESET Nod 32 - what do you guys think is better? And maybe Comodo Firewall.

Mate I am biased as everyone knows but if I were you Kaspersky 2012 ISS - the Pure is ok but gets complicated. Drop the WOT (see screen shot) as KIS does the same thing for you and best of all KIS has online banking. The firewall is just fine and it will switch of the Windows one anyway the defender needs disabling too the rest get rid of them you will not need them.
If you go down this track message me I'll give you a top flight config / setup for KIS.
You can run anything in a safe run if you want to but use it as a stand alone as it does NOT like other ISS's on with it.
I also run WPA2-PSK on my modem router.
The only one that is acceptable is the free Malwarebytes and then you need to have the scan settings right.

As far Trend micro is concern i feels it falls in category of mcafee and avg.
The last and best things which saves computer is "Images and Backup"MSE is OK for the "light" user. I had some problems with it and went back to Norton. That is a lot more robust. But I also do frequent images (3 times per week). So I can always bail mayself out.
You are right but Thanks i am not feeling any conflicts on machine.Hum GP that seems like an awful lot of overkill and well if you are not having any conflicts good luck but I am surprised you aren't.
I had uninstalled Emet and can only ditch zemana among other as no other software I can remove from machine.
Hitman pro had no impact and i tern off Mbam Real time and only use Realtime when i visit suspicious websites
Yes GP turning it off when not in use good move and there are some other good on call scanners besides Hitman especially if you suspect a rootkit or some other very undesirable creepy.
