The easiest way is probably "drag and drop". Open each disk in a separate window and drag the data from one disk to the other.

But it really depends on what kinds of drives you have (internal, external, spinner, solid state) and the type of data you're trying to move (large movies, small documents, encrypted, the operating system, etc.) Could you provide a screenshot of your Disk Management? And also fill in your System Specs.

How full is each drive? Is there room on either drive to fully contain the data from both drives? If so, move the data from one drive to the drive that has enough space to hold all of the data from both drives, which should leave one drive empty, then move the data you want from the drive containing all of the data to the empty drive.

Another possibility is to use a 3rd drive. Move data from drive E: to 3rd drive, move data from drive D: to drive E:, then move data from 3rd drive to drive D:.

In the time you have spent waiting for an answer to the fastest, best way, you could have easily copied the data using "Copy".

Windows has the command Robocopy built-in since Vista which should copy faster than the regular Copy command.

You could also look into the program Teracopy.

Quick way:

go into Disk Management
change the drive letter from "D" to "Z" (or any letter not used)
change the drive letter from "E" to "D".
change the drive letter from "Z" to "E".
That will change the drive letters from "D to E", and from "E to D" for those drives - quick and easy - no copy needed!
