Is this a data drive, with ONLY data on it?
If it is, I'd be tempted to move the data elsewhere temporarily.
Then reformat with NTFS and confirm the disk is OK with checkdisk or manufacturer's disk utilities.
Then move the data back.

Use a third party Partition Manager Boot Disk to convert from FAT to NTFS if you don't want to move the files already there. I think Windows 7 Disk Manager is not up to the task without deleting the data already on the disk.

Iggy beat me to it. If you answer the questions asked, perhaps we could be more specific.

At your own risk: try /r instead of /f with chkdsk (it's more thorough than /f).
It's not likely to hurt if your disk is healthy. And I've never lost data converting FAT to NTFS.
But, I always have proper backup, just in case. Cause you never know.

Are you sure you have entered the convert command in the proper syntax?
If you have correctly entered the command and it fails, I would try one or both of the following:
Use Partition Wizard, if I had my data backed up.
Reformat the drive, if I had my data backed up.
But I would do nothing at all if my data was not backed up. I wouldn't even use the convert command without a backup.
