I've never used tape to do backups before, but to the best of my knowledge, what you want to do cannot be done. Every drive has to have a different drive letter unless it is like WHS said you leave a blank drive letter which is next in line for windows to assign to it. But even then, if you plug something else in, it will have that letter by default. So, the software you are using will need to be the key. The software could be programed to back up to 1 drive on one day and the next drive the next day and so on and you would still get what you want. That is the only way I can think of to do it.
It is not the computer not remembering, it is the rules of the computer that each drive must have a different drive letter. That is the only way the computer distinguishes one drive from another.
Ok. Instead of having your script copy the files to a drive letter, would it be possible to have it copy to a drive name? I use FreeFileSync which does something similar to what your script does. It has an option to mirror files from folders (or a drive) to folders on another drive (or the drive itself). That second drive can be identified by the drive name instead of the drive letter. That way, it doesn't matter what the drive letter is as long as all the destination drives have the same drive name.

Why not use good old command line to assign Z: to the drive that is plugged in as a conditional command of there being a folder or file being there in the root. Try Take Command LE. That will solve your problem.
