I just read a flash drive used for Ready Boost should use exFAT file system.
I have a PNY 8 Gb in my laptop that I successfully reformatted from FAT32 to exFAT.
I have Patriot 8 Gb in one desktop that I tried to reformat from FAT32 to exFAT but it kept telling me it was in use. I rebooted that computer and tried again but failed. I moved it to another computer and tried again and failed.
I tried to delete the READY BOOST files and could not.
Why can't I reformat it?

You might need to use diskpart to clean the drive, before reformatting. You don't have your specs filled out, but if you have more than 1 GB of system memory, don't bother with ReadyBoost.

Have you disabled Ready Boost for the device?

Right click on the device in (My) Computer, open the Properties dialog box and disable Ready Boost. Safely remove the device, reboot your computer and plugin the drive. Check whether you can now delete the files in it and format.

Done!

After following the above instructions I finally changed the file to exFAT. I had to do jumanji's instructions twice before I could get it changed.
I was beginning to think I had a bad stick!
Thanks for everyones advice.
