I'm going to be upgrading my MB soon, when I remove my CPU from my current MB do I need to reapply any thermal paste or anything if I take the heatsink with it all in one? Or will I have to remove the heatsink and put the CPU in and then resink the cooler?

It would be best to remove the heatsink then remove the cpu individually.
Depending on the MB and heatsink, it will be inconsistent in seating the cpu in it's socket with the heatsink already attached.
If cpu not seated correctly, by by cpu, just to save on heatsink paste?
Depending on how long the cpu/heatsink have already been together, it would be worthwhile in replacing the heatsink anyway.
removing CPU and heatsink together is remarkably complex as the thermal paste isn't glue and most of the CPU-locking mechanisms in the socket are nearly impossible to operate with an heatsink mounted on it.
So yeah, do what they said, and use alcohol (the pink stuff) to clean both the CPU and the heatsink plate from any residue before applying new paste.

I often use Goo Gone or Goof Off to take off the old thermal paste. Then clean it with alcohol - I wouldn't want something shorting out in case the goo gone is conductive. The goo gone makes it SO much easier!
