No AV will prevent 100% of malware. Running a good AV with a good anti-malware program is a smart move. And just so people know, I run MBAM with real time protection enabled alongside NOD32. This has not caused any kind of conflict what-so-ever.
And btw Jimbo. In all the test results I've read (and that's a lot) MSE is mid-range at best. It is NOT "100% sufficient to do the job..." nor is it "as good or effective as 99.99% of other stuff" If you have reports/test results to support these claims, please provide a link.

A lot of those tests are essentially meaningless -- basically the "proof of the pudding is in the eating".
MSE is improving almost daily -- Magazine reports are often at least 30 and usually MORE days old so not up to date statistics either.
It really depends on how you use the Internet / email / social websites.
Some people will throughout their "whole computer lives" never experience a computer infection of any kind, others will attract infections like wasps around a jam jar whatever AV software is installed.
It really doesn't matter if AV product X can detect some really obscure Virus that AV product Y might miss if the user isn't likely to be logged on to the type of application or site that has this obscure virus.
In any case once the "obscure" virus is publicised it will become "less obscure" and definition updates will follow.
The worst that can happen is that you could (although if the virus was really obscure - it would be very unlikely that you would get infected in practice) get an infection for probably a maximum 24 hours at the most - assuming you scan at least every 24 hours and update daily -- so just restore back two days and you computer is clean again.
I would NEVER trust a computer that had been "cleansed" no matter HOW good the AV software is reputed to be -- a clean image restore IMO is the only sensible solution no matter what product you are using.
Note also that I'm referring here to HOME computers.
Protecting Corporate LANS or other "Multi User" computers is another subject entirely.
