Have a Sandisk 32 GB USB Flash Drive that I use for selective storage of certain files. If I purchase a Sandisk 64 GB to make a complete backup will I have to configure each one every time I swop usage. Why I ask this is that when I tried to introduce the new Sandisk to my laptop it could not be recognised as my computer was still configured to my old Verbatim 4 GB USB.
Your laptop should read both equally well at the same time.
Please insert both USB's into your laptop, and then post a maximised screen capture image of your Disk Management screen here so we can have a look at it for you:
1. Click rb:
2. Right-click My Computer
3. Click Manage
4. Click the Disk Management option
5. Maximise screen - take screen capture image and upload here

That really not necessary - you dont "configure" your computer to read different flash drives. Post the screen capture and we will have a look for you.
