First question here,
Is the other two USB ports work with other USB device?

If you're able to connect other device means it's functional.

I see it as a weird or unique situation here. :(
The most common problem with it is an external HDD consume more power source than any other USB devices, it needs very tight and secure connection on any available USB port on your machine to take 5 volts power input. DC power always rely on good contact between the port and the plug.

Some suggestion:
1. Inspect your External HDD cables for any dirt or stain.
2. Try checking the other two ports if it is a bit loose when is plugged.
3. Plug and hold in firm position for few more seconds to see if it works.
4. Consult with nearest ASUS local service to check the two USB voltage output.

That the only suggestions I can offer so far.

Hope it helps a little.

If this is one of the USB powered external drives more then likely it is not getting enough power from just one USB port to fully power it up.
In that case you will need a USB Y cable that connect to 2 USB ports at the same time to supply enough power to it.
