nowadays I am so much excited about encryption after watching BlackHat 2013 videos and Def Con 19 ,20

can u help me to find out words most powerful encryption software and methods

and where to learn it

I think doing PHD in Encryption will be best way for me

Truecrypt itself does real-time encryption/decryption when you mount and use its volumes. The data is really stored in the container file, and when you access the mounted drive, it decrypts on the fly in-memory, then re-encrypts when you save it back. Since it's open-source, you may take a look there to see how it's implemented in practice.
Windows itself has EFS and Bitlooker, that do something similar under the covers.
