I'm creating a usb with Tiny Core Live, can I use that to test the port?

Have never used that one so cannot say how it works .. I think your best option would be to Install Zorin instead of Windows

Either the Port or there is a fault in the Motherboard ....

And if it works on Linux is there anything I can do to get it working on Windows? I have another copy of Windows 7 that I can install.

Just restarted, and I think it installed because Windows suddenly started making the device connection sound
