Currently running Windows 7 Home Premium 64-bit
I need to upgrade to Windows 7 Professional 64-bit

Any pitfalls to be aware of?
Any suggestions for cheapest site to download iso?

Just open your Control Panel & select "Windows Anytime Upgrade".

I upgraded this way without a hitch, fairly quick process also.

If you are a student with a .edu email address you can get the student upgrade for $30. What the others said about it being a quick and painless upgrade was quite true, at least in my case.

Providing you've an upgrade key the Anytime Upgrade will work fine.

The main advice I'd offer is this:

Make sure you've downloaded and installed the latest updates first, otherwise you'll be in for a fairly lengthy wait while Windows downloads and installs updates before completing the upgrade.

If you upgrade, then install SP1, and then have to re-install for any reason make sure you carry out the upgrade again BEFORE you re-install SP1 or the upgrade will fail and you'll have to uninstall SP1 to carry out the anytime upgrade again.

When you've installed the upgrade, you'll need to activate again.

My advice would be to use the automatic telephone system by launching Start and type slui.exe 4 in the search field then press Enter. Choose your region for the correct telephone number and follow the automated instructions.

I always make a note of the installation ID first, and then write down and keep the activation ID before entering it in the activation ID fields.

That way, if you have to re-activate your upgrade for any reason just enter the activation ID without having to telephone first.

Alternatively, carry out a full image backup.

I would also capture a Win7 backup image before upgrade in case you ever want to revert to Home Premium and migrate your upgrade key elsewhere - all retail Win7 with the exception of builder's OEM can migrate for life.

Then save a Win7 backup image of the completed upgrade so you never have to reinstall either OS again.

anytime upgrade failed for me, I installed pro on my spare rig and then realised aero only works over remote desktop when the host is ultimate so tried to do a anytime upgrade to ultimate which failed with some long error code so I ended up clean installing ultimate.

You might want to try TeamViewer which I find is much more reliable and has better features than Remote Desktop.
